I've read two separate things, one says you check the trans level with the engine running and car in PARK, others say the car needs to be IN GEAR with the emergency brake on. Which is it? Does it matter?

Operating temperature in park. You need to run it through the gears and then in park and on a level surface. If you have a lift it makes it much easier since the car has to be level. A hot car on jack stands is a pain to get under. I suppose, if you had a pit, it would work nice too.

and by "run through the gears" you mean, P, R, N, and D?
